Effective Corneal Delivery of Moxifloxacin and Dexamethasone Combination Using Drug Eluting Mucoadhesive Contacts

Purpose: To design and develop drug eluting polymeric contacts for effective delivery of moxifloxacin (MF) and dexamethasone (DM).

Methods: Polymeric contact lenses were prepared using the polymer combination containing chitosan, glycerol and polyethylene glycol. Moxifloxacin and dexamethasone were loaded into the contacts both separately and in combination. The contacts were characterized for pH, tensile strength, thickness, swelling index, surface topography, FTIR, XRD, mucoadhesion, in vitro drug release kinetics, ex vivo human corneal permeation, in vivo bio-distribution in rabbit ocular tissues, and in vitro and in vivo antimicrobial efficacy. The drug-loaded contact lenses were compared with the standard drug solutions.

Results: Physical characteristics of polymeric contacts were similar to the commercially available contact lens. Cornea drug concentrations with MF loaded lens and MF std. were 91.48 ± 16.61 and 55.79 ± 6.75 µg/gram respectively. MF from the contacts showed sustained release up to 24h until which 80% drug was released DM loaded contacts showed release of >90% with in 24h. Ex vivo and in vivo antimicrobial activity of the MF loaded contacts were superior to the standard drug solutions.

Conclusion: Overall, MF and DM loaded polymeric contacts were efficient in delivering the drug at required therapeutic concentrations. These drug-eluting contact lenses could be used in the post-operative conditions to prevent ocular infection..
